ABSTRACT

Water magnetization and geoprocessing are increasingly utilized tools in weed management. Our objective was to study the influence of water magnetization on herbicide efficiency and to verify whether there is a relationship between control scores and the normalized difference vegetation index (NDVI). In the laboratory experiment, water was subjected to magnetization and evaluated with respect to four characteristics. In the field experiment, plots of Brachiaria grass were subjected to treatments in a factorial scheme (6 × 2 + 1). Six herbicidal factors (doses of glyphosate and glyphosate + 2,4-D) and the magnetization or absence of magnetization of the spray solution were evaluated and compared against the control treatment (without spraying). Weed control assessments were carried out six times. Images were obtained using an embedded multispectral camera to determine the NDVI values. Data related to water characteristics were analyzed using the t test. Weed control and NDVI data were subjected to analysis of variance and are presented in regression graphs. Dispersion analysis of NDVI data was performed according to the control scores. The magnetization process decreased the pH of the water and increased the surface tension, but it did not influence the control scores or the NDVI. As the glyphosate dose was increased, the control scores were higher and the NDVI values were lower. Magnetized water did not affect the biological efficiency of the herbicides, and there was a strong correlation between the control scores and the NDVI values.


A magnetização da água e o geoprocessamento são ferramentas cada vez mais utilizadas no manejo de ervas daninhas. Nosso objetivo foi estudar a influência da magnetização da água na eficiência do herbicida e verificar se existe uma relação entre os escores de controle e o índice de vegetação por diferença normalizada (NDVI). No experimento de laboratório, a água foi submetida à magnetização e avaliada em relação a quatro características. No experimento de campo, parcelas de capim-braquiária foram submetidas a tratamentos em esquema fatorial (6 × 2 + 1). Seis fatores herbicidas (doses de glyphosate e glyphosate + 2,4-D) e a magnetização ou ausência de magnetização da calda foram avaliados e comparados com o tratamento controle (sem pulverização). Avaliações de controle de plantas daninhas foram realizadas seis vezes. As imagens foram obtidas usando uma câmera multiespectral incorporada para determinar os valores de NDVI. Os dados relacionados às características da água foram analisados por meio do teste t. O controle de plantas daninhas e os dados de NDVI foram submetidos à análise de variância e são apresentados em gráficos de regressão. A análise de dispersão dos dados NDVI foi realizada de acordo com os escores de controle. O processo de magnetização diminuiu o pH da água e aumentou a tensão superficial, mas não influenciou nos escores de controle ou no NDVI. Conforme a dose de glyphosate foi aumentada, os escores de controle foram maiores e os valores de NDVI foram menores. A água magnetizada não afetou a eficiência biológica dos herbicidas, e houve uma forte correlação entre os escores de controle e os valores de NDVI.

ABSTRACT

Saflufenacil is a herbicide recommended for use in the main agricultural crops in Brazil to control eudicotyledonous weeds. However, the application of saflufenacil has been carried out without previous knowledge of this herbicide interactions with soil colloids, which may increase environmental contamination risks. In this study, the leaching of saflufenacil in soil samples was estimated with different organic matter contents. Therefore, PVC columns were filled with samples of Xanthic Ferralsol (XF) and Ferralsol (F), with different physical and chemical attributes. Twelve hours after herbicide (70 g i.a. ha-1) application, the PVC columns were subjected to a rainfall of 60 mm. For the evaluation of saflufenacil leaching, the columns were sectioned in ten parts with 5-cm each. Saflufenacil leached up to 50 and 40 cm deep in the samples, with the lowest contents of organic matter of XF and F, respectively. The increase in organic matter content of Ferralsol and Xanthic Ferralsol samples reduced saflufenacil leaching and the symptoms of intoxication in the indicator species. There was an inverse relation between the content of organic matter and the leaching of the herbicide in both soils studied. It was observed that the application of saflufenacil in soils with low organic matter contents may represent a significant environmental contamination risk of soils and watercourses.

Objectives: The herbicide glyphosate, a pesticide used in agriculture to control weeds, both in food crops and in other agricultural areas, has been identified as an endocrine modulator through the inhibition of aromatase activity and the activation of estrogen receptors. The present study examined the effects of a glyphosate-based herbicide (Roundup® (GLY-BH) on sexual dimorphism of rats after perinatal exposure to low and high GLY-BH in males and females offspring. Methods: Two groups of pregnant rats were treated with two doses of GLY-BH (50 or 150 mg/kg) from day 15 of gestation (GD15) to postnatal day 7 (PND7). Play fighting behavior was observed at the juvenile stage and during social and sexual behaviors in adulthood. Results: Perinatal GLY-BH exposure reduced male and female body weight at 28, 75, and 90 days of age. The play fighting behavior was decreased in both sexes, but female rats were more affected. The sexual behaviors were reduced only in females. Conclusions: Perinatal exposure to both doses of GLY-BH promoted sexually dimorphic effects in both juvenile and adulthood stages. These effects were attributed to the inhibition of aromatase activity induced by exposure to GLY-BH in the perinatal period.(AU)


Objetivos: O glifosato é um herbicida não seletivo, usado em muitas culturas alimentares e não alimentares e em áreas não agrícolas, sendo que os produtos a base de glifosato atuam como moduladores das funções endócrinas por meio da inibição da atividade da aromatase e da ativação de receptores de estrógeno. O presente estudo avaliou os efeitos do herbicida Roundup® (GLY-BH) à base de glifosato, em comportamentos sexualmente dimórficos de ratos após exposição perinatal a doses baixas e altas de GLY-BH no período perinatal. Métodos: Ratas prenhas foram tratadas com 50 ou 150 mg/kg de GLY-BH do 15º dia de gestação (GD15) ao 7º dia de lactação (LD7). O comportamento de luta/brincar foi observado na fase juvenil e os comportamentos social e sexual na idade adulta. Resultados: a exposição perinatal a GLY-BH reduziu o peso corporal de machos e fêmeas aos 28, 75 e 90 dias de idade. O comportamento de luta/brincar diminuiu em ambos os sexos, sendo as ratas foram as mais afetadas. O comportamento sexual foi reduzido apenas nas fêmeas. Conclusões: A exposição perinatal a ambas as doses do GLY- BH promoveu tanto na idade juvenil como na idade adulta, efeitos sexualmente dimórficos. Esses efeitos foram atribuídos à inibição da atividade da aromatase induzida exposição perinatal ao GLY-BH.(AU)

ABSTRACT

Abstract To discover and isolate a glyphosate-resistant gene from Fragaria vesca through gene mining. An open reading frame (ORF) of 1563 bp encoding EPSPSwas amplified from Fragaria vesca (FvEPSPS). FvEPSPS (Genebank: XP004306932.1) encodes a polypeptide of 520 amino acids and it has hightly homologous with EPSPS from other plants. qRT-PCR analysis showed that the FvEPSPS was expressed extensively in all tissues including leaves, roots and stems, with higher expression in leaves. Furthermore, transgenic Arabidopsis Thaliana exhibited 10 mM glyphosate to resistance. Therefore, this research offers a new glyphosate-resistant gene for development of transgenic crops.

Production and storage environments influence the physiological and sanitary quality of seeds. This study aimed to evaluate the effects of the herbicide flumioxazin applied as a preharvest desiccant and storage on the physiological and sanitary quality of adzuki bean seeds. A randomized block design was used in a split-plot scheme, with four replications. Plots consisted of five doses of the herbicide flumioxazin (20, 25, 30, 35, and 40 g ai·ha-1) and subplots consisted of two evaluation times (harvest and six months after harvest). Two controls, with no herbicide application, were tested for each evaluation time. The physiological seed quality was maintained with doses from 25 to 35 g·ha-1 of flumioxazin. Increasing herbicide doses reduced the electrical conductivity of seeds. The increased doses also reduced the percentage of normal seedlings in the first germination count after six months of storage and caused seedling vigor loss. The storage of adzuki bean for six months reduced its physiological quality, leading to losses in germination and seed vigor. Storage and increasing doses of the desiccant flumioxazin reduced the incidence of the pathogens Rhizopus, Cladosporium, and Fusarium in the seeds. The increased doses of this herbicide reduced Aspergillus incidence. Penicillium incidence was not affected by treatments.(AU)


A qualidade fisiológica e sanitária de sementes é influenciada pelo ambiente de produção e de armazenamento. Neste trabalho objetivou-se avaliar os efeitos do herbicida flumioxazin aplicado como dessecante em pré-colheita e do armazenamento sobre a qualidade fisiológica e sanitária de sementes de feijão azuki. Foi adotado o delineamento experimental em blocos ao acaso, em esquema de parcelas subdivididas. Nas parcelas foram alocadas as cinco doses do herbicida flumioxazin (20, 25, 30, 35 e 40 g i.a·ha-1) e nas subparcelas as duas épocas de avaliação (colheita e 6 meses após a colheita), com quatro repetições. Foram testados dois tratamentos sem aplicação do herbicida para cada época de avaliação. Nas doses entre 25 a 35 g·ha-1 de flumioxazin observou-se a manutenção da qualidade fisiológica das sementes. Em doses crescentes reduziu-se a condutividade elétrica das sementes. O aumento das doses reduziu a porcentagem de plântulas normais na primeira contagem da germinação, após seis meses de armazenamento, e causou perda no vigor das plântulas. O armazenamento de feijão azuki durante seis meses reduziu a qualidade fisiológica, causando perdas na germinação e vigor das sementes. O armazenamento e o aumento das doses do dessecante flumioxazin reduziram a incidência dos patógenos Rhizopus, Cladosporium e Fusarium nas sementes. O aumento das doses reduziu a incidência de Aspergillus. A incidência de Penicillium não foi afetada pelos tratamentos.(AU)

ABSTRACT

Brazil is the worldwide leader in the long-term use of pesticides and herbicides. This compromises the health of handlers by causing harmful neurological, respiratory, and cardiovascular changes. The herbicide 2.4D has been shown to cause cardiac overload with subsequent pathological remodeling. Objective: To analyze the cardiac morphological repercussions on the left ventricle (LV) of mice submitted to nebulization by the herbicide 2.4D. Methods: Fifteen mice were divided into three groups: control group (CG; n = 5) exposed to nebulization with sodium chloride solution; low concentration group (LCG; n = 5) exposed to nebulization of the herbicide 2.4D with 3.71 x 10-3 grams; and high concentration group (HCG; n = 5) exposed to nebulization of the herbicide 2.4D with 9.28 x 10-3 grams for 15 minutes. The fractal dimension analysis was performed through the box-counting method. Later, the ImageJ program was used to calculate the fractal dimension of each group. To evaluate cardiac remodeling, histological slides were prepared and stained with Hematoxylin-Eosin (HE). Fifty areas of cardiomyocytes were analyzed per animal. The comparisons between groups were performed by ANOVA One-Way with Tukey's posttest (p < 0.05). Results: There was no change in fractal dimension values between the CG = 1.37 ± 0.02, LCG = 1.33 ± 0.04 and the HCG = 1.33 ± 0.07 groups. However, cardiac hypertrophy occurred in the HCG = 303.9 ± 38.80 µm(2) when compared to the CG group = 236.9 ± 61.71 µm(2) (p = 0.034). Conclusion: The herbicide 2.4D used for 72 hours did not promote cardiotoxicity when evaluated by fractal dimension. However, cardiomyocyte hypertrophy was observed in the LV

Abstract In Brazil, the expansion of agricultural activity and the associated indiscriminate use of herbicides such as glyphosate is directly related to the loss of biodiversity in the Cerrado. The identification of plant species as bioindicators of herbicide action, especially species native to the area, can help in monitoring the impacts of xenobiotics in the remaining Cerrado. Thus, this study was designed to evaluate the possible use of the native Cerrado species Pouteria torta as a bioindicator of glyphosate action via changes in physiological performance. At 16 months after sowing, the effect of glyphosate was evaluated by applying the following doses: 0 (control), 25, 50, 100, 200, 400, 800, and 1200 g a.e. ha-1. In response to glyphosate, P. torta exhibited reductions in photosynthesis and chloroplastid pigment content, as well as accumulation of shikimic acid and the occurrence of chlorosis and necrosis. These changes demonstrate the high sensitivity of P. torta to glyphosate and its potential for use as a bioindicator of this herbicide.


Resumo No Brasil, a expansão da atividade agrícola, aliada a utilização indiscriminada de herbicidas como o glyphosate, possui relação direta com a perda da biodiversidade no Cerrado. A identificação de espécies vegetais bioindicadoras da ação de herbicidas, particularmente as nativas do Cerrado, pode auxiliar em processos de monitoramento dos impactos desse xenobiótico nas remanescentes do Cerrado. Assim, este estudo foi projetado para avaliar o possível uso de Pouteria torta, espécie nativa do cerrado, como bioindicadora da ação do glyphosate via mudanças na sua performance fisiológica. Após 16 meses de semeadura, o efeito do glyphosate foi avaliado quando aplicadas as seguintes doses: 0 (controle), 25, 50, 100, 200, 400, 800 e 1200 g e. a. ha-1. Em reposta ao glyphosate, as plantas de P. torta apresentaram redução na sua performance do processo fotossintético e no conteúdo de pigmentos cloroplastídicos, além do acúmulo de ácido chiquímico e da ocorrência de cloroses e necroses. Essas alterações demonstram a alta sensibilidade de P. torta ao glyphosate, o que potencializa a sua utilização como bioindicadora da ação desse herbicida.

ABSTRACT

O uso inadequado de herbicidas pode resultar em intoxicações agudas e, às vezes, crônicas por exposição em longo prazo a baixos níveis desses agentes tóxicos, podendo o herbicida atuar também como agentes teratogênicos, mutagênicos, cancerígenos e desreguladores endócrinos, com o aparecimento de doenças neurodegenerativas e distúrbios reprodutivos. Estudos têm revelado que a melatonina tem propriedades antioxidantes, anti-inflamatórias e imunomoduladoras e atua na reprodução. Essa indolamina está entre os agentes que têm se mostrado benéfico em intoxicações por herbicidas, porém não há relatos do uso de melatonina contra intoxicações por Glifosato-Roundup®, muito menos em associação com o Paraquat. Dessa forma, o maior interesse no tratamento das intoxicações por herbicidas, tem-se concentrado em medidas que impeçam ou minimizem as lesões celulares provocadas nos diversos sistemas biológicos. Assim, a melatonina, como antioxidante conhecido, pode ser mais uma alternativa contra as intoxicações por herbicidas associados e/ou individuais.


The inadequate use of herbicides may cause serious and sometimes chronic poisoning due to long exposure to low levels of toxic agents. Herbicides may also be teratogenic, mutagenic, cancerigenous agents and endocrine disruptors, with the occurrence of neurodegenerative diseases and reproduction disorders. Several studies have shown that melatonin has antioxidant, anti-inflammatory and immune-modulating qualities, besides affecting the reproduction system. It is among the agents which are beneficent in poisoning by herbicides even though no reports are extant on the use of melatonin against poisoning by Glyphosate-Roundup® alone or associated with Paraquat. Solutions that prevent or minimize cell lesions caused by several biological systems have been focused upon in the treatment for poisoning with herbicides. Thus, melatonin, a known antioxidant, may be an alternative against the poisoning by single or associated herbicides.

ABSTRACT

Rhamdia quelen (silver catfish) and Leporinus obtusidens (piava) were exposed to a commercial formulation Roundup(r), a glyphosate-based herbicide at concentrations of 0.2 or 0.4 mg/L for 96 h. The effects of the herbicide were analyzed on the alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities and glucose in plasma, glucose and protein in the mucus layer, nucleotide hydrolysis in the brain, and protein carbonyl in the liver. The parameters were chosen, owing to a lack of information concerning integrated analysis, considering oxidative damage parameters, liver damage, and effects on the mucus layer composition and triphosphate diphosphohydrolase (NTPDase) activities. Plasmatic glucose levels were reduced in both species, whereas the transaminase activities (ALT and AST) increased after exposure to the herbicide. Herbicide exposure increased protein and glucose levels in the mucus layer in both species. There was a reduction in both NTPDase and ecto-5'-nucleotidase activity in the brain of piava, and increased enzyme activity in silver catfish at both concentrations tested. The species showed an increase in protein carbonyl in the liver after exposure to both concentrations of the glyphosate. Our results demonstrated that exposure to Roundup(r) caused liver damage, as evidenced by increased plasma transaminases and liver protein carbonyl in both of the fish species studied. The mucus composition changed and hypoglycemia was detected after Roundup(r) exposure in both species. Brain nucleotide hydrolysis showed a different response for each fish species studied. These parameters indicated some important and potential indicators of glyphosate contamination in aquatic ecosystems.

Os efeitos do herbicida foram analisados na atividade da alanina aminotransferase (ALT), aspartato aminotransferase (AST) e glicose no plasma, glicose e proteína na camada de muco, hidrólise de nucleotídeos no cérebro e a proteína carbonil no fígado. Os parâmetros foram escolhidos devido à falta de informação com relação a análises integradas, considerando parâmetros oxidativo, danos no fígado, efeitos na composição da camada do muco e atividade da trifosfato difosfoidrolase (NTPDase). Níveis de glicose plasmática foram reduzidos em ambas às espécies, enquanto a atividade das transaminases (ALT e AST) aumentou após exposição ao herbicida. A exposição ao herbicida aumentou a proteína e níveis de glicose na camada de muco em ambas as espécies. Houve uma redução em ambas atividades de NTPDase e ecto-5'-nucleotidase no cérebro de piava, e um aumentou a atividade destas enzimas em jundiás em ambas as concentrações testadas. As espécies mostraram um aumento na proteína carbonil no fígado após exposição a ambas as concentrações de glifosato. Nossos resultados demonstraram que a exposição ao Roundup(r) causou danos no fígado, como evidenciado pelo aumento das transaminases plasmáticas e proteína carbonil no fígado em ambas as espécies de peixes estudadas. A composição do muco alterou e uma hipoglicemia foi detectada após a exposição ao Roundup(r) em ambas espécies. A hidrólise de nucleotídeos em cérebro mostrou diferente resposta para cada espécie estudada. Esses parâmetros indicam alguns importantes e indicadores potenciais da contaminação do glifosato no ecossistema aquático.

ABSTRACT

The 2,4-dichlorophenoxyacetic acid, usually named 2,4-D is one of the most widely used herbicides in the world. Acute toxicity of 2,4-D herbicide was investigated through its effects on guppies (Poecilia vivipara Bloch et Schneider 1801). Fish were exposed to the herbicide at concentrations of 10, 20 and 40µl per liter of water for 24 hours to determine its effects on gills and liver epithelia. The estimated LC50 was 34.64µl of 2,4-D per liter of water. Histochemical analyses and Feulgen's reaction were conducted to detect glycoconjugates and DNA, respectively, in gills and liver epithelia. Histochemistry revealed qualitative variations of glycoconjugates present on mucous cells and granules. The four types of mucous cells contained neutral granules, acids, or both. Increasing amounts of syalomucins were observed from the control group to the group exposed to the highest concentration of 2,4-D, suggesting increased mucous viscosity and the formation of plaques that could inhibit gas exchange and osmoregulation. Lamellar fusion observed in the group exposed to 40µl of 2,4-D suggests a defense mechanism. Hepatocytes showed vacuolization in the 10 and 20µl/L groups. The 40 µl/L group showed normal hepatocytes as well as changed ones, many Ito cells, micronuclei, and nuclear swelling. These effects may be associated with toxicity or adaptative processes to cellular stress. The data from this study indicates the importance of assessing similar risks to aquatic species and suggests that Poecilia vivipara is an adequate biological model for analysis of environmental contamination.


A toxicidade aguda do herbicida 2,4-D foi investigada através dos efeitos no peixe Poecilia vivípara (Bloch et Schneider, 1801). Grupos de peixes foram expostos ao herbicida nas concentrações de 10, 20 e 40µl por litro de água, durante 24 horas. As brânquias e o fígado foram estudados. A concentração letal média (CL50) do herbicida para a espécie em questão foi de 34,64µl/l. Foram realizadas colorações histoquímicas e coloração de Feulgen para identificar glicoconjugados e DNA, respectivamente, nos tecidos acima citados. Os métodos histoquímicos revelaram os tipos de glicoconjugados presentes nas células mucosas e nos grânulos. Os quatro tipos de células mucosas apresentaram glicoconjugados neutros, ácidos, ou ambos em um mesmo tipo celular. Observou-se a presença crescente de sialomucinas do grupo controle até o grupo exposto a maior concentração de 2,4-D, sugerindo aumento da viscosidade do muco e, consequentemente, formação de placas que impedem as trocas gasosas e a osmorregulação. A fusão lamelar observada no grupo exposto a 40µl de 2,4-D sugere ser um mecanismo de defesa. Os hepatócitos apresentaram processo de vacuolização nos grupos 10 e 20µl/l. No grupo de 40µl/l, observou-se a presença de células de Ito, micronúcleos e hepatócitos normais e outros com edema nuclear. Este estudo indica a importância da avaliação de riscos semelhantes a espécies aquáticas e sugere a espécie Poecilia vivipara como modelo biológico adequado para análises de contaminação ambiental.

ABSTRACT

Cattle can be considered as an important source for herbicides through nutrition. Therefore, herbicide residue in animal products is a potential human exposure to herbicides causing public health problems in human life. Triazines are a group of herbicides primarily used to control broadleaf weeds in corn and other feed ingredients and are considered as possible human carcinogens. To evaluate trace residue of these pollutants molecular imprinted solid phase extraction [MISPE] method has been developed, using biological samples. Blood samples were taken from the jugular vein of 45 Holstein cows in 3 commercial dairy farms in Khuzestan Province, Iran. Urine samples were also taken from the cows. The mean +/- SD concentrations of atrazine in serum and urine samples of the study group [0.739 +/- 0.567 ppm and 1.389 +/- 0.633 ppm, respectively] were higher [P < 0.05] than the concentrations in serum and urine samples of the control group [0.002 +/- 0.005 ppm and 0.012 +/- 0.026 ppm, respectively]. Atrazine in the feed ingredients ingested by cattle could be transferred into the biological samples and consequently can be considered as a potential hazard for the public health

ABSTRACT

Glyphosate [N-(phosphonomethyl)-glycine] is a broad-spectrum, non-selective, post-emergence herbicide that is extensively used in agriculture. Published data referring to the effects of this product on human health are contradictory. We showed previously that long-term treatment of rats with low doses of Glyphosate-Biocarb® may induce hepatic histological changes and bleeding without decreasing platelet counts. The aim of the current study was to investigate, in vitro, the effect of glyphosate on human blood platelet aggregation and coagulation. Materials and methods: Platelet aggregation was determined in the platelet-rich plasma using the agents: 6µM-adenosine diphosphate, 6µM-epinephrine and 4µg/mL-collagen. Pretreatment with 500µg/mL glyphosate showed significant hypofunction of the three aggregating agents. The inhibitory effect was dose-dependent at concentrations from 50 to 500 µg/mL. The release of ATP was lower for glyphosate-treated platelets after stimulation by collagen. On the other hand, glyphosate did not promote any inhibitory effects on prothrombin time, thromboplastin time and thrombin time. In conclusion, the results demonstrate that glyphosate promotes changes in the platelet metabolism with an inhibitory effect on primary hemostasis.


O glifosato [N-(phosphonomethyl)-glycine] é um herbicida pós-emergente não seletivo de amplo espectro muito utilizado na agricultura. Dados da literatura referentes aos efeitos desse produto na saúde humana são contraditórios. Em estudos prévios demonstramos que ratos previamente tratados com glifosato apresentavam lesões hepáticas e sangramento sem alterações quantitativas de plaquetas. O objetivo do presente estudo é investigar os efeitos in vitro do glifosato (GP) na agregação plaquetária e coagulação sanguínea em humanos. A agregação plaquetária foi determinada em plasma rico em plaquetas (PRP) usando os agentes adenosina difosfato (ADP) 6µM, epinefrina 6µM e colágeno 4µg/mL. Pré-tratamento com GP 500µg/mL demonstrou significativa hipofunção dos três agentes agregantes. O efeito inibitório foi dose dependente em concentrações de 50-500 µg/mL. Utilizando-se a quantificação de ATP como um índice da capacidade de secreção plaquetária, foi observado diminuição da liberação das plaquetas tratadas com GP. Por outro lado, o GP não promoveu efeito inibidor no tempo de protrombina (TP), tempo de tromboplastina parcial ativada (ATTP) e tempo de trombina (TT). Em conclusão, os resultados demonstram que o GP promove mudanças no metabolismo plaquetário com efeito inibitório na hemostasia primária.

ABSTRACT

The present study was undertaken to assess the environmental hazards presented to wildlife by new pesticides or new uses of established pesticides, biochemical changes, residual behavior and accumulation of diclofop-methyl [Iloxan 36 % EC]. A laboratory trials of diclofop-methyl were carried out on tilapia fish, Oreochromis niloticus as indicator species to . Acute toxicity of diclofop-methyl [Iloxan 36 % EC] was carried out and the median lethal concentration for 96 hours [96 h - LC50] value was 1.89 mg / l. In the present study, two groups of fish were exposed to two sub lethal concentrations selected on the basis of 1 / 10 [0.19 mg / l, low concentration] and 1 / 3 [0.63 mg / l, high concentration] of diclofop-methyl LC50 value for 28 days under the laboratory conditions compared to the control group. The treatments indicated that a gradual bioaccumulation of herbicide with time in whole fish was more pronounced than in muscle. Several criteria for estimating pesticide accumulation effect were studied on different biochemical parameters in plasma of fish after treatments. The results indicated that alanine aminotransferases [ALT], alkaline phosphatase [ALP] and acetylcholinesterase [AChE] activities as well as albumin and glucose levels increased significantly after treatment with both concentrations comparing to control group. However, significant increase in aspartate aminotransferases [AST] and total protein levels with was recorded low concentration [0.19 mg / l] only. On the other hand, high concentration [0.63mg / l] caused significant increase in urea levels. However, ceriatinine level was decreased in both exposed concentrations. Noticeable changes in antioxidants biomarkers were recorded, a reduction in SH-protein level accompanied with an elevation in lipid peroxidation biomarker [MDA] were recorded in plasma after treatment with two concentrations. The previous results were accompanied with noticeable alterations in plasma and muscle protein patterns in both treated groups

ABSTRACT

Descreve-se um método de isolamento de mitocôndrias acopladas de tilápia-do-nilo Oreochromis niloticus, isoladas de células hepáticas de peixes adultos. As mitocôndrias estavam metabolicamente ativas, sendo capazes de realizarem fosforilação oxidativa, de acordo com os valores do quociente de controle respiratório. Os valores de controle respiratório obtidos com malato/piruvato (complexo I) e com succinato (complexo II) foram de 5,8±0,8 e 3,38±0,4, respectivamente. O potencial de membrana exibiu o valor de 197±4mV, quer se utilizasse malato/piruvato ou succinato como substrato. O procedimento de isolamento de mitocôndrias de O. niloticus permite o estudo do efeito de xenobióticos na bioenergética mitocondrial, tendo sido avaliada a ação da oxifluorfena (0,6mgL-1) na bioenergética mitocondrial. Os resultados demonstram que o tratamento com oxifluorfena influencia a capacidade fosforilativa dos peixes, interferindo na sua carga energética, o que poderá levar à sua morte.


A method for isolation of coupled mitochondria isolated from the liver of adult Nile tilapia Oreochromis niloticus is described for the first time. They were metabolically active, able to sustain oxidative phosphorylation, as shown by respiratory control ratio values, which were about 5.8±0.8 and 3.3±0.4 when respiring on malate/piruvate (complex I) or succinate (complex II), respectively, as substrate. Membrane potential exhibited a value of approximately 197±4mV for malate/piruvate or succinate. The procedure now described for the isolation of O. niloticus mitochondria is an important new tool, allowing the study about the effect of xenobiotics on mitochondrial bioenergetic, being evaluated the effect of oxyfluorfen (0.6mgL-1) in the liver mitocondrial bioenergetic. These results showed that phosphorylation was significantly affected by oxyfluorfen which contributed to the decrease on the liver cell energy charge and consequently led to the fish dead.

ABSTRACT

O Glifosato é um herbicida não-seletivo, sistêmico e suas vendas representam em torno de 60% das vendas de herbicida em todo o planeta, totalizando uma quantia de 1,2 bilhões de dólares anuais. Este princípio ativo é apontado como um potencial contaminante ambiental, juntamente a seu metabólito, o ácido aminometilfosfônico (AMPA), visto que são altamente hidrossolúveis. A degradação de Glifosato em AMPA ocorre rapidamente no meio ambiente. A dispersão pelo ar e a contaminação de corpos hídricos, são potenciais fontes de exposição a seres humanos, que pode acontecer de forma direta ou indireta, sendo que a exposição crônica a estas substâncias vem sendo estudada com resultados que sugerem correlação entre a exposição e desregulação endócrina, câncer e dermatites. O objetivo deste trabalho foi avaliar o processo de contaminação ambiental por Glifosato, na área rural da Serrinha do Mendanha, pertencente ao bairro de Campo Grande, município do Rio de Janeiro. Essa comunidade é considerada como periurbana, apesar das características comuns às comunidades agrícolas, devido à proximidade de centros urbanos. O local é caracterizado pela presença de pequenas propriedades, onde as principais culturas são as de chuchu, abobrinha, banana, aipim, jiló e quiabo, observando-se um elevado consumo do herbicida Glifosato. Para a avaliação da contaminação ambiental foi otimizado um método de análise para o Glifosato em água. Além disso, foram avaliadas as diferenças no processo de trabalho da área periurbana em relação a áreas exclusivamente rurais. The herbicide glyphosate is a non-selective, systemic, and its sales represent around 60% of thesales of herbicide around the world, totaling an amount of 1.2 billion dollars annually. This active principle is identified as a potential environmental contaminant, together with its metabolite, the aminomethylphosphonic acid (AMPA), since they are highly water soluble. The degradation ofglyphosate in AMPA occurs rapidly in the environment. The scattering by air and contamination of water bodies are potential sources of exposure to humans, which can happen directly or indirectly, Chronic exposure to these substances has been studied with results that suggest a correlation between exposure and endocrine disruption, cancer and dermatitis. The objective of this study was to evaluate the process of environmental contamination by Glyphosate in the rural area of Serrinha do Mendanha which belongs to the neighborhood of Campo Grande, Rio de Janeiro. This community is considered periurban due to the proximity to urban centers,despite the common characteristics with other agriculture communities. The site is characterized by the presence of small properties, where the main crops are vegetables, observing a high consumption of the herbicide glyphosate. To assess environmental contamination an analytical method was optimized to determine glyphosate in water. ABSTRACT

Acute toxicity of the glyphosate -N (phosphonomethyl) glycine- herbicide, Roundup®, in juveniles of cachama blanca, (Piaractus brachypomus), was evaluated and the histopathological lesions were assessed. The 96 h lethal concentration 50 was 97.47mg.L-1 (P<0.05). In the gill, necrotic and proliferative lesions were detected. In the liver, congestion, degenerative foci, hyaline droplets and lipidic vacuolization of the hepatocytes were observed. In the stomach mild hyperplasia of mucous cells was detected, which was also observed in the skin. In this latter tissue, a large increase in the thickness of the epidermis with necrotic lesions, infiltration of leukocytes and melanin pigment were observed. In the brain, degenerative foci of neuronal bodies in the telencephalon associated with gliosis and infiltration of eosinophilic granule cells/mast cells were shown. In conclusion, gills, liver, skin and brain are susceptible to Roundup®. Moreover, effects on the central nervous system could affect olfaction as well as individual and group behavior, the reproductive performance of the fish and hence have repercussions at the population level.


Este estudo avaliou a toxicidade aguda e as alterações histopatológicas induzidas pelo glifosato -N (fosfometil) glicina, na formulação Roundup® em juvenis de pirapitinga (Piaractus brachypomus) expostos durante 96 horas. A concentração letal 50 foi de 97.47mg.L-1 (P<0.05). Nas brânquias foram achadas lesões proliferativas e necróticas. No fígado foi observada congestão, processos degenerativos, gotas hialinas e presença de vacúolos lipídicos nos hepatócitos. No estomago e na pele foi detectada hiperplasia ligeira das células de moco. Nesta ultima, adicionalmente foi observado engrossamento da epidermes com lesões necróticas, infiltração de células leucocitárias e acumulação de melanina. No cérebro foram observados focos de degeneração de somas neurais na região do telencéfalo junto com gliose e infiltração de células granulais eosinofilicas/células mast. Concluindo, as brânquias, o fígado, a pele e o cérebro são órgãos suscetíveis ao Roundup®. Adicionalmente, os efeitos sobre o sistema nervoso central poderiam reduzir a olfação nos peixes, tanto como o comportamento grupal e individual, a performance reprodutivo e desta forma, repercutir no nível populacional.

ABSTRACT

Os efeitos causados pelas substâncias contidas nos pesticidas e adjuvantes podem ser responsáveis por inúmeras alterações no sistema reprodutivo de machos e de fêmeas. A partir do momento que o glifosato-Roundup penetra na célula ele reduz a atividade da proteína StAR e da enzima aromatase. Este trabalho avaliou a possível correlação entre o desenvolvimento das características puberais em animais expostos diariamente ao herbicida glifosato, pela observação do crescimento e desenvolvimento e do início do período púbere. Utilizou-se 32 ratos machos divididos em 4 grupos de tratamentos (0, 5, 50 e 250 mg/kgPV), dos 23 aos 53 dias de idade. Utilizou-se análise de MANCOVA para a comparação dos pesos corporais, Kruskall-Wallis para o dia e ANOVA para o peso ao descolamento do prepúcio. Não houve interferências do tratamento sobre o crescimento dos animais em nenhum grupo. A idade à puberdade foi significativamente diferente entre os grupos 50 mg/kg e 0 mg/kg (36,6±0,5; 36 dias; p<0,05), 250 mg/kg e 0 mg/kg (37,2±0,4; 36 dias; p<0,001) e 5 mg/kg e 250 mg/kg (36,5±0,53; 36 dias; p<0,05). O peso à puberdade foi maior no grupo de 250 mg/kg em relação aos grupos de 0 mg/kg e 5 mg/kg (142,7 ± 9,3; 128,9 ± 5,4; 126,1 ± 8,8 g, respectivamente; p<0,001). O peso do grupo de 50 mg/kg (134,1 ± 9,2g) não foi diferente dos outros grupos. É provável que a exposição crônica ao herbicida glifosato-Roundup cause a disrupção endócrina no eixo hipotalâmico-hipofisário-gonadal durante a maturação sexual, pela evidenciação do atraso no início da puberdade.


The effects of herbicides and it's adjuvants can be responsible for several alterations in reproductive organs of male and female. Since glyphosate-Roundup penetrates in the cell, it reduces the activity of StAR protein and aromatase enzime. This study evaluated the development of pubertal characteristics in animals receiving oral dosages of glyphosate-Roundup by gavage, by daily body weight measurement and verification of preputial separation (PPS). 32 male rats were allocated in 4 groups of treatments (0, 5, 50 e 250 mg/kg) from 23 to 53 days of age. The body weight from 23 to 53 days was compared using analysis of MANCOVA, Kruskall-Wallis was used to compare the day at PPS and ANOVA was used to compare the body weight at PPS. Age at PPS was different between groups 50 mg/kg and 0 mg/kg (36,6±0,5; 36 days; p<0,05), 250 mg/kg and 0 mg/kg (37,2±0,4; 36 days; p<0,001) and 5 mg/kg and 250 mg/kg (36,5±0,5; 36 days; p<0,05). The body weight at PPS was higher in group of 250 mg/kg n comparison to groups of 0 mg/kg and 5 mg/kg (142,7 ± 9,3; 128,9 ± 5,4; 126,1 ± 8,8 g, respectively; p<0,001). The body weight of group 50 mg/kg (134,1 ± 9,2g) wasn't differ from another groups. Probably, the daily exposure to glyphosate-Roundup causes endocrine disruption in hypothalamic-pituitary-gonad axis during sexual maturation, because it delayed the onset of puberty.

ABSTRACT

Glyphosate is the active ingredient in the most largely used herbicide in the world and has been held responsible, in scientific studies, for toxic effects in cases of accute intoxication through ingestion in suicide attempts. Toxic effects caused by occupational exposures to different glyphosate formulations have not been indentified. The existing commercial glyphosate formulations have several ingredients that are called adjuvants wich, nevertheless, have toxicological effects and may cause distinct symptoms. The objetive of this work was to review the cases of occupational exposure to formulations of glyphosate and the toxic effects found in farm workers who were examined at the Toxicology Ambulatory at Unicamp. From a total of 334 patients, 160 of them were exposed to fromulations of glyphosate and 146 presented distinct clinical signs and symptons with the same kind of exposure. We have reached the conclusion that adjuvants components in the formulations may be rsponsible for more important toxicological effects than active ingredient, glyphosate...

ABSTRACT

Present study was carried out to examine the effect of Butachlor on growth and nitrogen fixation by Anabaena sphaerica. The increased concentration of the pesticide did not have any adverse effect on the alga. Rather it accelerated the algal contribution in terms of biomass and nitrogen fixation.
